A Miniature Culture System for Optimization of Chlorella vulgaris Cultivation

Introduction
High-throughput cultivation and screening methods allow a parallel, miniaturized, and cost efficient processing of many samples. These methods however have not been generally established for phototrophic organisms such as filamentous or spiral microalgae.
In this study, a microplate system was used for screening of Chlorella vulgaris as a unicellular micro-algal strain, for best mass production by different Carbon, Nitrogen and phosphate concentration in the medium.

Method
Pure cultures of microalgae strains such as Chlorella vulgaris were cultivated in 250ml flasks and 24-well microplates with different concentrations of nitrate and phosphate. All growth experiments were conducted in BBM medium, and cultures were put on shaker with the rate of 150 rpm under the fluorescent light source. Cell density was determined by microscope count method using a hemocytometer and optical density was measured by Nano-Drop spectrophotometer at 590 nm wavelength.

Results
Similarity percentage of cultures was about 85% and correlation ratio of Chlorella growth in BBM medium between different amounts of Carbon, Nitrogen and phosphor source (C:N:P) was obtained, about 90:14:1.

Conclusion
With this miniature culture system, the effects of different C, N and P sources and initial pH, were examined on Chlorella growth and biomass production. Also, there was good agreement between Micro-plate and usual culture methods, such as flask culture on shaker.
The microplate data were comparable to similar data obtained in flask and photo-bioreactors.
